Sus antecedentes:

  • La alergia alimentaria implica respuestas inmunes perjudiciales de tipo 2, pero los mecanismos de tolerancia no están claros.
  • La tolerancia oral a los antígenos de la dieta es crucial para prevenir la FA y la anafilaxia.
  • La molécula beta similar a la resistina (RELMβ) derivada de las células cápsulas está implicada en la regulación inmune.

Objetivo del estudio:

  • Investigar el papel de RELMβ en la regulación de la tolerancia oral a los antígenos de los alimentos.
  • Aclarar los mecanismos por los que RELMβ influye en la tolerancia a los alimentos y en el desarrollo de la FA.
  • Identificar posibles objetivos terapéuticos para la prevención y el tratamiento de la FA.

Principales métodos:

  • Se utilizaron modelos de ratón de FA y estudios de deleción génica (RELMβ knockout).
  • Análisis de los niveles séricos de RELMβ en pacientes con FA y en modelos de ratón.
  • Se ha investigado el impacto de la RELMβ en la composición y función del microbioma intestinal (metabolismo indólico).
  • Se evaluó el papel de los derivados de indol y las células T reguladoras de RORγt+ (Treg) en la tolerancia.
  • Se evaluó el potencial terapéutico del antagonismo de RELMβ durante el período peri-destete.

Principales resultados:

  • La RELMβ está elevada en pacientes con FA y en modelos de ratón.
  • Los ratones protegidos por la deleción de RELMβ de FA, la producción de IgE y la anafilaxia.
  • RELMβ interrumpió la tolerancia oral por el agotamiento de Lactobacilli y Alistipes productores de indol.
  • Los derivados del indole promueven las células RORγt+ Treg protectoras de la FA a través de la activación del receptor de hidrocarburos de arilo.
  • El antagonismo de RELMβ durante el destete restauró la tolerancia y evitó la FA en ratones susceptibles.

Conclusiones:

  • RELMβ es un regulador crítico de la tolerancia oral, mediando un circuito inmuno-epitelial intestinal.
  • RELMβ funciona a través de la edición del microbioma, impactando el control innato de la inmunidad adaptativa.
  • Dirigirse al eje RELMβ-microbioma-indole-Treg ofrece una nueva estrategia para la prevención y el tratamiento de la FA.
