Sus antecedentes:

  • Los neutrófilos son conocidos principalmente por sus funciones proinflamatorias y el aclaramiento de patógenos.
  • Algunos subconjuntos de neutrófilos poseen propiedades inmunosupresoras, pero sus mecanismos no se comprenden completamente.

Objetivo del estudio:

  • Investigar las posibles funciones antiinflamatorias de los neutrófilos más allá del aclaramiento del patógeno.
  • Caracterizar las nuevas vesículas extracelulares producidas por neutrófilos envejecidos.

Principales métodos:

  • Caracterización de las grandes vesículas derivadas de neutrófilos de envejecimiento (LAND-V) basadas en el tamaño, la estructura y la biogénesis.
  • Análisis de las señales de "no me comas" en los LAND-V y su interacción con los fagocitos.
  • Investigación del papel de CD55 en los efectos antiinflamatorios mediados por LAND-V.
  • Identificación de los dominios de la balsa lipídica y las vías dependientes de RhoA en la formación de LAND-V.

Principales resultados:

  • Los neutrófilos producen vesículas extracelulares antiinflamatorias únicas (LAND-V) que difieren de los tipos de vesículas clásicas.
  • Los LAND-V exhiben señales de "no me comas", evitando la fagocitosis y acumulándose durante la resolución de la inflamación.
  • CD55 en LAND-Vs inhibe la conversión del complemento C3, reduciendo el reclutamiento de neutrófilos y el daño tisular.
  • Los LAND-V se originan en balsas de lípidos en neutrófilos envejecidos a través de brotes dependientes de RhoA.

Conclusiones:

  • Los LAND-V son nuevos inmunomoduladores producidos por neutrófilos que resuelven activamente la inflamación.
  • Los LAND-V que expresan CD55 proporcionan efectos antiinflamatorios sostenidos y mitigan el daño tisular.
  • Los LAND-V representan un objetivo terapéutico potencial para enfermedades inflamatorias e infecciosas.
