Sus antecedentes:

  • El corazón en desarrollo de los mamíferos depende de la comunicación entre el endocardio y el miocardio, separados por gelatina cardíaca.
  • Los mecanismos de comunicación intercelular a través de la gelatina cardíaca durante la morfogénesis cardíaca siguen sin estar claros.

Objetivo del estudio:

  • Investigar cómo los receptores y ligandos localizados en la membrana median las interacciones entre el endocardio y el miocardio durante el desarrollo cardíaco.
  • Aclarar el papel de las microestructuras específicas en la facilitación de esta comunicación.

Principales métodos:

  • Utilizado en el desarrollo de modelos de morfogénesis cardíaca de ratón.
  • Empleó una combinación de enfoques celulares, de imágenes y genéticos.
  • Investigó la estructura y la función de las conexiones intercelulares.

Principales resultados:

  • Se han identificado estructuras de túneles similares a nanotubos (TNTL) que median la interacción directa entre los cardiomiocitos (CM) y las células endocárdicas (EC).
  • Se demostró que las TNTL transportan proteínas citoplasmáticas y transducen señales entre las CM y las EC.
  • Se han observado TNTL que inician el crecimiento del miocardio, contribuyendo a la formación de trabéculas ventriculares.

Conclusiones:

  • Las TNTL son un nuevo mecanismo para la comunicación intercelular directa a través de la gelatina cardíaca en el corazón en desarrollo.
  • La interrupción de las TNTL afecta las interacciones de señalización esenciales, lo que lleva a defectos en el patrón ventricular.
  • Estos hallazgos proporcionan nuevos conocimientos sobre la base celular de la morfogénesis cardíaca.
