Resumen
Los nuevos métodos irCLIP-RNP y Re-CLIP revelan ensamblajes dinámicos de ARN-proteína. Estas técnicas mapean las interacciones de proteínas que se unen al ARN y sus roles en los procesos celulares y las enfermedades, avanzando en nuestra comprensión de la regulación génica.

Principales métodos:
- Desarrollo y aplicación de la ligadura no isotópica inducida por la luz ultravioleta y la inmunoprecipitación combinada con la espectrometría de masas (irCLIP-RNP).
- Utilizando irCLIP-RNP para identificar las RDAP coligadas al ARN con las RBP específicas.
- Desarrollo de un método de inmunoprecipitación secuencial irCLIP (Re-CLIP) para identificar ARN unidos por múltiples RBP simultáneamente.
Principales resultados:
- irCLIP-RNP definió con éxito paisajes de ensamblajes de proteínas multiméricas en el ARN, revelando patrones de asociación RBP-ARN selectivos por tipo de célula.
- Se observó una remodelación dinámica de los RDAP en respuesta al EGF, con el reclutamiento de UPF1 cerca del HNRNPC promoviendo la supervisión de empalme de los ARNm de proliferación.
- Re-CLIP confirmó los hallazgos de irCLIP-RNP e identificó la unión conjunta de HNRNPC y UPF1 en los ARNm RND3 y DDX3X.
