Objetivo del estudio:

  • Introducir PathoPlex, un marco escalable, controlado por calidad e interpretable para imágenes altamente multiplexadas con resolución subcelular.
  • Desarrollar un paquete de software para extraer e interpretar patrones de coexpresión de proteínas a través de capas biológicas.
  • Demostrar la utilidad de PathoPlex en la identificación de características patológicas y objetivos terapéuticos en la enfermedad renal y la diabetes.

Principales métodos:

  • PathoPlex combina imágenes altamente multiplexadas (más de 140 anticuerpos a 80 nm / píxel, 95 ciclos) con un software especializado.
  • El marco fue optimizado para el procesamiento de muestras de biopsia de archivo, permitiendo el análisis simultáneo de al menos 40 muestras.
  • Se extrajeron y analizaron patrones de coexpresión de proteínas (agrupaciones) a través de dominios celulares y subcelulares.

Principales resultados:

  • PathoPlex identificó la actividad epitelial de JUN como un factor clave en la enfermedad renal mediada por el sistema inmunológico.
  • El análisis de la enfermedad renal diabética humana relacionó los grupos de proteínas a nivel del paciente con la disfunción de los órganos e identificó el estrés tubular mediado por el calcio como objetivo terapéutico.
  • Se identificaron agrupaciones relacionadas con el estrés renal en individuos con diabetes tipo 2 sin enfermedad renal histológica, y se evaluaron las respuestas a los inhibidores de SGLT2.

Conclusiones:

  • PathoPlex proporciona una solución potente y escalable para el análisis integrador de imágenes en tejidos complejos.
  • El marco facilita la democratización de las imágenes multiplexadas, lo que permite una visión más profunda de los mecanismos de la enfermedad.
  • PathoPlex apoya el desarrollo de atlas de patología de nueva generación y ayuda a identificar nuevas estrategias terapéuticas.
