Sus antecedentes:

  • El síndrome del ventrículo único e hipoplástico del corazón izquierdo (SV/HLHS) plantea riesgos significativos para múltiples sistemas de órganos, incluida la función inmune.
  • Los pacientes con SV/HLHS presentan sistemas inmunológicos debilitados, aumentando la susceptibilidad a infecciones y complicaciones cardiovasculares.

Objetivo del estudio:

  • Investigar el impacto de la SV/HLHS en las células mononucleares de la sangre periférica (PBMC) a nivel de una sola célula.
  • Identificar tipos específicos de células inmunes y vías moleculares afectadas por el SV/HLHS.

Principales métodos:

  • Se empleó secuenciación de ARN unicelular (scRNA-seq) para analizar las PBMC de 33 pacientes pediátricos con SV/ HLHS y 33 controles sanos.
  • El análisis ponderado de la red de coexpresión génica (WGCNA) se utilizó para identificar patrones clave de expresión génica y diferencias específicas de tipo celular.

Principales resultados:

  • Cuatro tipos clave de células Th1/17, TFH, NK y Th2 mostraron los cambios más significativos en la expresión génica relacionados con la enfermedad.
  • Un total de 6.659 genes fueron expresados diferencialmente, con notables diferencias de sexo en las células TFH.
  • Se encontró que la actividad de MYC e IFN-γ estaban sobrerrepresentadas en las células T y NK, y subrepresentadas en los monocitos y las células Treg.

Conclusiones:

  • El estudio revela un complejo panorama de transcriptomas de células inmunes en pacientes con SV/HLHS.
  • Comprender la modulación de la actividad de MYC e IFN-γ ofrece objetivos terapéuticos potenciales para equilibrar las respuestas inmunes, reducir la inflamación y mejorar la reparación de los tejidos.
