Resumen

El envejecimiento saludable está relacionado con un desgaste más lento de los telómeros y menos defectos de reparación del ADN. Los factores genéticos, incluida la reducción de la carga de variantes germinales de alto impacto funcional y la mayor resistencia al estrés oxidativo, contribuyen a un envejecimiento más saludable y al bienestar en la vida posterior.

Sus antecedentes:

  • La inestabilidad genómica, el deterioro de la reparación del ADN y el desgaste de los telómeros son factores clave del envejecimiento.
  • Las diferencias en estos factores entre el envejecimiento saludable y el no saludable siguen sin estar claras.

Objetivo del estudio:

  • Investigar las diferencias genómicas y moleculares entre el envejecimiento saludable y no saludable.
  • Identificar los factores genéticos que contribuyen al envejecimiento saludable.
  • Explorar el papel de la longitud de los telómeros, las variantes de la línea germinal y el estrés oxidativo en el envejecimiento.

Principales métodos:

  • Secuenciación del genoma completo (WGS) en 100 individuos envejecidos sanos y 100 no sanos.
  • Medición de la longitud de los telómeros mediante TelSeq y Computel.
  • Análisis de la carga de variantes germinales de alto impacto funcional (gHFI), alteraciones de la vía y conjunto de datos GTEx para el impacto funcional.

Principales resultados:

  • La longitud de los telómeros disminuyó rápidamente en los ancianos enfermos después de los 65 años.
  • Los ancianos sanos exhibieron una menor carga de gHFI, especialmente en los genes de reparación del ADN (por ejemplo, BLM).
  • El análisis de la vía mostró un enriquecimiento de las mutaciones de estrés oxidativo en personas mayores sanas, correlacionadas con un estrés oxidativo reducido y enzimas antioxidantes reguladas al alza (SOD1, SOD2).

Conclusiones:

  • La estabilidad genómica preservada a través del lento desgaste de los telómeros y la reducción de los defectos de reparación del ADN es clave para un envejecimiento saludable.
  • Una mayor resistencia al estrés oxidativo puede promover un envejecimiento más saludable.
  • Los factores genéticos juegan un papel importante en la mitigación del deterioro relacionado con la edad y el apoyo al bienestar.
