Stem cells are undifferentiated cells that divide and produce different cell types. Resumen

Las células madre embrionarias humanas RSeT (hESC) representan un estado pluripotente distinto, diferente de los estados ingenuos y primados. Estas células muestran propiedades de crecimiento únicas y carecen de marcadores de pluripotencia ingenuos clave, ofreciendo nuevas perspectivas sobre las transiciones de estado de las células madre.

Sus antecedentes:

  • Las células madre embrionarias humanas (hESC) exhiben estados pluripotentes ingenuos y primados.
  • Trabajos anteriores identificaron estados pluripotentes ingenuos heterogéneos de varios protocolos.
  • Comprender la heterogeneidad del estado pluripotente es crucial para las aplicaciones de células madre.

Objetivo del estudio:

  • Para caracterizar un estado pluripotente comercial basado en RSeT.
  • Investigar el comportamiento de la RSeT hESC en diferentes condiciones de crecimiento.
  • Definir las propiedades transcriptómicas y bioquímicas de las RSeT hESC en relación con otros estados pluripotentes.

Principales métodos:

  • Cultivo celular bajo normoxia e hipoxia.
  • Ensayos de eficiencia del revestimiento de una sola célula.
  • Análisis integrador del transcriptoma.
  • Análisis de la expresión del marcador superficial (SUSD2, CD75).
  • Ensayos bioquímicos para la dependencia de las vías de señalización (FGF2, JAK, TGFβ).

Principales resultados:

  • Las hESC RSeT pueden crecer sin hipoxia, pero muestran un crecimiento y una eficiencia de recubrimiento variables.
  • Las RSeT hPSC carecen de las características transcriptómicas de la pluripotencia ingenua y formativa.
  • Las hESC RSeT se asemejan a los embriones posteriores a la implantación temprana, similares a las hESC primarias.
  • Los marcadores de superficie ingenuos SUSD2 y CD75 no se expresan de manera significativa.
  • Las hESC de RSeT exhiben dependencia específica de la línea celular de FGF2 y co-independencia de la señalización de JAK/TGFβ.

Conclusiones:

  • Las RSeT hESC representan un nuevo estado pluripotente aguas abajo de la pluripotencia ingenua.
  • El medio RSeT puede restringir el potencial pluripotente ingenuo manteniendo la actividad de FGF2.
  • Este estudio mejora la comprensión de las transiciones de estado pluripotente in vitro.
