Sus antecedentes:
- El sistema hipotalámico de hipocretina / orexina (Hcrt / Ox) es crucial para regular los ciclos de sueño-vigilia, y su disfunción está relacionada con la narcolepsia.
- Los niveles bajos de Hcrt/Ox en el líquido cefalorraquídeo (LCR) son un biomarcador específico para la narcolepsia, sin embargo, la neurotransmisión del sistema sigue sin estar clara.
Objetivo del estudio:
- Para investigar la transmisión sináptica frente al volumen de péptidos Hcrt/Ox.
- Determinar el papel de los receptores Hcrt/Ox en la liberación y síntesis de péptidos.
- Para explorar el dimorfismo sexual dentro del sistema Hcrt/Ox.
Principales métodos:
- Microscopía electrónica del locus coeruleus de la rata para analizar la distribución de las vesículas Hcrt/Ox.
- La administración intraperitoneal de suvorexant (un antagonista dual de los receptores Hcrt/ Ox) a las ratas.
- Prueba de inmunosorbción ligada a enzimas (ELISA) para cuantificar los niveles de Hcrt/Ox en el LCR y los sinaptosomas.
Principales resultados:
- La liberación de Hcrt/Ox-péptido ocurre predominantemente a través de la transmisión volumétrica/extrasináptica en lugar de la liberación sináptica.
