Leaves can be only millimeters in width or tens of meters wide, depending on the environment. Objetivo del estudio:

  • Para aclarar la base molecular de la determinación de la forma de la hoja en la col china.
  • Para identificar el gen responsable de la formación de hojas lobuladas.

Principales métodos:

  • La clonación basada en el mapa para identificar el locus del rasgo cuantitativo (QTL) qBrLLA10.
  • Análisis de la secuencia de alelos parentales y del gen BrRCO.
  • La sobreexpresión ectópica en Arabidopsis y el silenciamiento genético inducido por virus (VIGS) en B. rapa.
  • Análisis sintético en las adhesiones de Brassica rapa.

Principales resultados:

  • Identificó BrRCO, un factor de transcripción HD-Zip, como el gen causal para la formación de hojas lobuladas.
  • Las variaciones de la región promotora en BrRCO explican las diferencias de forma de la hoja entre las líneas progenitoras.
  • BrRCO regula positivamente la formación del lóbulo de la hoja; su regulación a la baja bloquea el desarrollo del lóbulo.
  • Las regiones promotoras altamente variables y las regiones de codificación conservadas de BrRCO sugieren que la evolución cis-regulatoria impulsa la diversificación funcional.

Conclusiones:

  • BrRCO es un regulador positivo de la formación de los lóbulos de las hojas en Brassica rapa.
  • Las modificaciones cis-reguladoras en el promotor de BrRCO son responsables de la variación funcional y la diversidad de la forma de la hoja.
  • Los hallazgos proporcionan información para mejorar la forma de la hoja en la col china y otras especies de Brassica.
