Este estudio identifica seis polimorfismos de un solo nucleótido (SNPs) altamente patógenos en el gen PRKCQ, relacionados con el cáncer. Estas variantes afectan la estabilidad y función de la proteína quinasa C theta (PKCθ), lo que sugiere nuevos objetivos terapéuticos.

Sus antecedentes:

  • Los polimorfismos de un solo nucleótido (SNP) son clave para la variación genética individual, la evolución y la susceptibilidad a las enfermedades, incluidos el cáncer y la diabetes.
  • El gen PRKCQ codifica la proteína quinasa C theta (PKCθ), una enzima crucial en las respuestas inmunes y el desarrollo del cáncer.

Objetivo del estudio:

  • Identificar y caracterizar los SNPs no sinónimos patógenos (nsSNPs) en el gen PRKCQ.
  • Evaluar el impacto de estos NSNP en la estructura, la estabilidad y la función de la proteína PKCθ.
  • Explorar las implicaciones de estos hallazgos para las enfermedades asociadas con PRKCQ y la medicina personalizada.

Principales métodos:

  • Utilizó múltiples herramientas de bioinformática para la evaluación de la patogenicidad de los NSNP.
  • Realizó predicciones estructurales, análisis de dominio y perfiles de conservación de PKCθ.

Principales resultados:

  • Se han identificado seis nsSNP altamente patógenos y oncogénicos en el PRKCQ (rs1838691533 R6W, rs145984477 P27L, rs1248923790 C29Y, rs1837738907 R145C, rs1837738573 R146W, rs1403981107 L495P).
  • Se encontraron variantes ubicadas en dominios funcionales críticos de PKCθ, que afectan la unión a la membrana y la actividad catalítica.
  • Se demostró que la mayoría de los nsSNP disminuyen la estabilidad de las proteínas y alteran significativamente la conformación y la dinámica de PKCθ.

Conclusiones:

  • Los nsSNP patógenos en PRKCQ pueden afectar críticamente la función de PKCθ, contribuyendo a la señalización oncogénica.
  • Estos hallazgos ponen de relieve el papel de PRKCQ en el cáncer y sugieren el potencial de terapias dirigidas.
  • El estudio apoya los enfoques de medicina personalizada para las enfermedades asociadas con las variaciones de PRKCQ.
