Sus antecedentes:

  • El adenocarcinoma ductal pancreático (PDAC) presenta niveles elevados de marcadores de oxidación de ácidos grasos peroxisomales (FAO) en comparación con los individuos sanos.
  • Peroxisomal FAO está implicado en la progresión de la PDAC, lo que requiere investigación en modelos preclínicos.

Objetivo del estudio:

  • Investigar el papel de la acetilcoenzima A aciltransferasa 1 (ACAA1) en la FAO peroxisomal dentro del PDAC.
  • Evaluar el potencial terapéutico de dirigirse a ACAA1 en modelos de PDAC.

Principales métodos:

  • Se generaron ratones knockout ACAA1 y se cruzaron con ratones KPC (modelo PDAC de ingeniería genética) para evaluar la supervivencia.
  • Se utilizó un modelo de xenotransplante de ratón con knockdown de ACAA1 en células PDAC humanas para evaluar el crecimiento tumoral y los cambios metabólicos.
  • Se midió el consumo de oxígeno, la producción de ATP y los marcadores de autofagia (LC3-II) en respuesta a la eliminación de ACAA1.

Principales resultados:

  • La eliminación de ACAA1 redujo significativamente el consumo de oxígeno (hasta un 60%) y la producción de ATP (hasta un 70%) en las células PDAC, pero no en las células normales.
  • La desactivación de ACAA1 indujo la autofagia (aumento de los niveles de LC3-II) y la inactivación de mTOR en las células PDAC, lo que condujo a un retraso en el crecimiento en modelos de xenotransplante.
  • Los ratones heterocigotos ACAA1 cruzados con ratones KPC mostraron un aumento de la supervivencia media de 3 semanas.

Conclusiones:

  • Dirigirse a ACAA1 inhibe el crecimiento del tumor PDAC al inducir la autofagia, que apoya la supervivencia de los ratones modelo PDAC.
  • La inhibición de ACAA1 se dirige específicamente al metabolismo energético de las células cancerosas, ofreciendo una estrategia terapéutica potencial con un impacto mínimo en las células normales.
