Sus antecedentes:

  • Los metabolitos del triptófano (Trp) son cruciales para la inmunidad antitumoral y la respuesta a la inmunoterapia.
  • El papel preciso de Trp en los linfocitos T citotóxicos específicos del tumor (CTL) dentro del microambiente tumoral sigue sin estar claro.

Objetivo del estudio:

  • Aclarar el mecanismo del metabolismo de Trp en las CTL activadas por el tumor y su impacto en la inmunidad antitumoral.
  • Investigar el eje Slc7a5-Trp-AhR-FasL en la función CTL y su potencial terapéutico.

Principales métodos:

  • Metabolomía de todo el genoma, secuenciación de ARN y secuenciación de ATAC en CTL y células tumorales específicas co-cultivadas.
  • Inhibición farmacológica de Slc7a5 y modelos de deficiencia genética en ratones.
  • Secuenciación de scRNA, inmunoprecipitación de cromatina y experimentos de bloqueo de FasL.
  • Análisis de datos de pacientes humanos con cáncer que correlacionan la expresión génica con la respuesta a la inmunoterapia y la supervivencia.

Principales resultados:

  • Los niveles de Trp se elevaron en las CTL activadas por el tumor, que aumentaron la expresión de Slc7a5.
  • La inhibición de Slc7a5 redujo la absorción de Trp y la actividad lítica de CTL.
  • La deficiencia de Slc7a5 específica de las células T alteró la inmunidad antitumoral, aumentando el crecimiento tumoral y la metástasis.
  • La deficiencia de Slc7a5 disminuyó la activación del receptor de hidrocarburos de arilo (AhR) y la expresión de FasL en las células T.
  • AhR se une directamente con el promotor de Faslg.
  • El bloqueo de FasL exacerbó la progresión del tumor.
  • En los cánceres humanos, la expresión de AhR y FasL se correlacionó, con FasL relacionado con la respuesta y la supervivencia al pembrolizumab.

Conclusiones:

  • La vía metabólica Slc7a5-Trp es esencial para la activación de AhR, que regula el FasL en las células T que se infiltran en el tumor, manteniendo así la inmunidad antitumoral CTL.
  • Dirigirse a Slc7a5 para mejorar la función de las células T presenta una estrategia prometedora para la inmunoterapia del cáncer, mejorando potencialmente la eficacia de las células CAR-T.
