Resumen

Sus antecedentes:

Objetivo del estudio:

  • Realizar un estudio de aleatorización mendelano bidireccional para evaluar la posible relación causal entre el estreñimiento y la EP.
  • Para determinar si el estreñimiento es una característica prodroma de la EP o si la EP influye en el estreñimiento.
  • Para utilizar las variantes genéticas para inferir la causalidad entre estas condiciones.

Principales métodos:

  • Se utilizó un diseño de estudio de aleatorización mendeliana bidireccional de 2 muestras.
  • Las variantes genéticas asociadas con el estreñimiento se obtuvieron del consorcio FinnGen.
  • Para analizar la asociación bidireccional se utilizaron los datos resumidos del estudio de asociación de todo el genoma para la EP.

Principales resultados:

  • No se encontró ningún efecto causal estadísticamente significativo entre el estreñimiento y la EP en ninguna de las direcciones.
  • El análisis anticipado (efecto del estreñimiento en la EP) no mostró una asociación significativa (OR=0,77, P=0,097).
  • El análisis inverso (efecto de la DP en el estreñimiento) tampoco reveló un vínculo causal significativo (OR=1,00, P=0,845).

Conclusiones:

  • El estudio no encontró una asociación genética significativa entre el estreñimiento y la enfermedad de Parkinson en las poblaciones europeas.
  • Estos hallazgos desafían la hipótesis del estreñimiento como una característica prodromica temprana de la EP.
  • Se necesita más investigación con cohortes más grandes y multiétnicas y ajuste covariado.
