Sus antecedentes:

  • Las nanopartículas de upconversion dopadas con lantánidos (UCNPs) ofrecen fotostabilidad y bajo fondo para la bioimagen.
  • El brillo limitado de una sola partícula restringe su amplia aplicación en estudios biológicos.
  • La mejora de la migración de energía (EM) y la eficiencia de la transferencia es crucial para las UCNPs más brillantes.

Objetivo del estudio:

  • Superar el brillo limitado de las UCNP para mejorar las aplicaciones de bioimagen.
  • Mejorar la migración de energía (EM) entre los sensibilizadores Yb y los emisores Er.
  • Desarrollar UCNPs con un brillo de una sola partícula superior para estudios de transporte neuronal.

Principales métodos:

  • UCNP diseñados para inhibir la transferencia de energía hacia atrás (BET).
  • Aumento del dopaje de Yb3+ en el núcleo (NaLu0.9-xYbxEr0.1F4@NaYbF4@NaLuF4) para aumentar el EM.
  • Utilizó el modelo de Markov oculto bayesiano para el análisis cuantitativo de la dinámica de transporte neuronal.

Principales resultados:

  • Los UCNPs con un núcleo de aleación (NaYb0.9Er0.1F4) mostraron un aumento de más de diez veces en la luminiscencia de conversión hacia arriba.
  • La relación Yb3+/Er3+ optimizada y el grosor de la cáscara inerte maximizaron el brillo de una sola partícula.
  • Habilitado el seguimiento a largo plazo del transporte axonal en las neuronas del ganglio de la raíz dorsal (DRG).

Conclusiones:

  • Los UCNP optimizados mejoran significativamente el brillo de una sola partícula a través de una mejor migración de energía.
  • Reveló un mecanismo de coordinación cinesin-dinina en el transporte neuronal.
  • Establecido el seguimiento de una sola partícula de conversión ascendente (uSPT) como una herramienta poderosa para el monitoreo de la actividad neuronal en tiempo real.
