Sus antecedentes:

  • El cambio de fenotipo permite a las células cancerosas alternar entre estados proliferativos e invasivos.
  • Las señales externas que desencadenan esta plasticidad celular permanecen en gran medida sin identificar.

Objetivo del estudio:

  • Investigar el papel del confinamiento mecánico en la mediación del cambio de fenotipo de las células cancerosas.
  • Identificar los mecanismos moleculares subyacentes a los cambios de fenotipo inducidos por el confinamiento.

Principales métodos:

  • Utilizó un modelo de melanoma de pez cebra y muestras humanas.
  • Empleó transcriptómica espacial y de una sola célula para perfilar las células tumorales.
  • Análisis morfológico y modelado cuantitativo.

Principales resultados:

  • El confinamiento mecánico induce la remodelación de la cromatina, lo que lleva al cambio de fenotipo.
  • HMGB2 identificado como un mediador clave, regulado en células confinadas.
  • Las células de melanoma con alto contenido de HMGB2 presentan una proliferación reducida y una mayor resistencia a los fármacos.

Conclusiones:

  • El microambiente mecánico es un conductor crítico del cambio de fenotipo del melanoma.
  • HMGB2 media el intercambio entre los estados proliferativos e invasivos.
  • La orientación de las señales mecánicas o HMGB2 puede ofrecer nuevas estrategias terapéuticas.
