Sus antecedentes:

  • Los ARN circulares (ARNc) y los ARN mensajeros (ARNm) se originan a partir de los mismos genes, pero se producen a través de mecanismos de empalme distintos.
  • Este estudio investiga el comportamiento comparativo de transcriptomas circulares y lineales en diversas condiciones y tejidos biológicos.

Objetivo del estudio:

  • Analizar las distintas lógicas reguladoras de los transcriptomas circulares y lineales.
  • Comprender sus funciones en respuesta a la infección y durante el desarrollo de los tejidos y la variación fisiológica.

Principales métodos:

  • Análisis de los datos transcriptómicos de muestras de macrófagos derivados de monocitos bovinos durante el estudio de Mycobacterium avium ssp. infección por paratuberculosis (MAP).
  • Examen de datos transcriptómicos de tejidos bovinos sanos (testículos, hígado, músculo) estratificados por edad, sexo y eficiencia alimentaria.

Principales resultados:

  • El transcriptoma circular es aproximadamente 100 veces más pequeño que el transcriptoma lineal.
  • La infección por MAP afectó principalmente al transcriptoma lineal en el ganado susceptible (JD-), mientras que los transcriptomas circulares mostraron respuestas menos específicas.
  • La maduración de los tejidos y los factores fisiológicos como el sexo y la eficiencia de la alimentación indujeron cambios distintos, a menudo no coordinados, en los transcriptomas circulares y lineales.

Conclusiones:

  • Los transcriptomas circulares y lineales operan bajo mecanismos regulatorios en gran medida independientes.
  • Los CircRNA pueden exhibir una mayor variabilidad y potencialmente llevar menos señales fisiológicas directas en comparación con los mRNA, especialmente en ausencia de firmas transcriptómicas lineales claras.
  • El análisis paralelo de ambos circRNAs y mRNAs es esencial para una comprensión completa de la regulación génica.
