Sus antecedentes:

  • Las herramientas de edición de genoma multiplexadas enfrentan desafíos en el procesamiento del ARNCR precursor (pre-crRNA) y requieren componentes regulatorios adicionales.
  • La nucleasa Cas12i.3 carece de capacidad de procesamiento pre-crRNA, lo que limita su potencial de multiplexión.

Objetivo del estudio:

  • Diseñar un sistema de edición del genoma multiplexado altamente eficiente y simplificado basado en Cas12i.3.
  • Para superar las limitaciones del tipo salvaje Cas12i.3 para aplicaciones amplias de edición del genoma.

Principales métodos:

  • Diseño optimizado de ARN CRISPR (crRNA), uso de codones y fusión de exonucleasa para crear Cas12i optimizado inicial (IOCas12i).
  • Diseño racional empleado y mutaciones de aminoácidos para desarrollar sistemas mejorados y optimizados de Cas12i (EOCas12i-Combo1 y EOCas12i-Combo2).
  • Evaluación de la eficiencia de edición, la especificidad y la capacidad de multiplexación utilizando hasta 30 matrices de ARNcr objetivo.

Principales resultados:

  • Los sistemas EOCas12i diseñados demostraron eficiencias de edición significativamente mejoradas (de 2,5 a 60,0 veces) en comparación con el Cas12i3 de tipo salvaje.
  • Se han logrado eficiencias de edición comparables a los sistemas establecidos como Streptococcus pyogenes Cas9 (SpCas9) y la bacteria Lachnospiraceae Cas12a (LbCas12a).
  • Se ha demostrado una edición multiplexada eficiente de hasta 30 objetivos utilizando matrices compactas de crRNA, produciendo indels más largos beneficiosos para el nocaut genético.

Conclusiones:

  • Los sistemas EOCas12i-Combo1 y EOCas12i-Combo2 desarrollados representan avances significativos en la edición multiplexada del genoma.
  • Estas nucleasas diseñadas ofrecen una plataforma simplificada y altamente eficiente para diversas aplicaciones de edición de genomas.
  • Las variantes mejoradas de Cas12i superan las limitaciones anteriores, allanando el camino para un uso más amplio en la investigación y la ingeniería genética.
