El ADN ribosómico nuclear (nrDNA) en las especies de morera (Morus) muestra una evolución concertada incompleta, con un alto polimorfismo en la región del espaciador transcrito interno (ITS). Este hallazgo ayuda en la taxonomía y cría de Morus.

Sus antecedentes:

  • El espaciador transcrito interno (ITS) es crucial para la taxonomía del género Morus, pero las características del ADN ribosomal nuclear (nrDNA) en todo el genoma no se comprenden bien.
  • Si bien la evolución de ITS generalmente es concertada, existen excepciones, lo que sugiere complejidades potenciales en la evolución de nrDNA dentro de Morus.

Objetivo del estudio:

  • Para investigar las características del ADNr de todo el genoma en el género Morus.
  • Evaluar el grado de evolución concertada del ADNnr en las especies de Morus.
  • Desarrollar nuevos marcadores moleculares para la identificación de especies de Morus y el análisis filogenético.

Principales métodos:

  • Identificación y análisis de polimorfismos de un solo nucleótido (SNPs) e inserciones/deleciones (InDels) dentro de las regiones de ADN nr de 542 accesos de morera.
  • Validación del polimorfismo ITS mediante clonación y secuenciación de Sanger.
  • Desarrollo de un marcador de secuencia polimórfica amplificada (CAPS) para la diferenciación de especies.

Principales resultados:

  • Se identificaron 158 SNPs y 15 InDels en dieciséis especies de Morus, revelando un polimorfismo intraindividual significativo y una evolución concertada incompleta de nrDNA.
  • Las regiones ITS (ITS1-5.8S-ITS2) mostraron un alto polimorfismo, con 66 SNPs y 13 InDels.
  • Se desarrolló un nuevo marcador de CAPS utilizando un 13/16 bp InDel en ITS1, lo que permite una rápida discriminación de M. alba y M. notabilis.

Conclusiones:

  • El ADN nr en Morus exhibe una evolución concertada incompleta, con un sustancial polimorfismo intraindividual, particularmente en las regiones ITS.
  • El marcador CAPS desarrollado proporciona una herramienta eficiente para distinguir las especies clave de Morus, evitando la secuenciación tradicional o el análisis fenotípico.
  • Los hallazgos mejoran la comprensión de la taxonomía de Morus, apoyando los esfuerzos de conservación y los programas de cría de la morera.
