Sus antecedentes:

  • Investiga el secukinumab (SECU) y el honokiol (HONK) en un modelo murino de asma alérgica con lesión pulmonar aguda (ALI).
  • Se centra en la modulación de las vías de citoquinas Th17 y Th1/Th2.
  • Aborda la necesidad de nuevas estrategias terapéuticas para los trastornos inflamatorios respiratorios complejos.

Objetivo del estudio:

  • Evaluar el potencial inmunomodulador de SECU y HONK, solos y en combinación.
  • Evaluar los efectos sinérgicos sobre los mediadores inflamatorios en un modelo de asma concomitante y ILA.
  • Para explorar los efectos antiinflamatorios adicionales de HONK en la producción de citoquinas.

Principales métodos:

  • Ratones sensibilizados y provocados con ovalbúmina (OVA) y lipopolisacárido (LPS).
  • La administración de SECU, HONK o terapia combinada.
  • Análisis cuantitativo de IgE, recuento de células BALF, perfiles de citoquinas e histopatología.

Principales resultados:

  • Tanto SECU como HONK demostraron efectos inmunomoduladores significativos.
  • La terapia combinada redujo sinérgicamente los mediadores proinflamatorios, incluidos el TNF-α y la IL-6.
  • Mejoras observadas en la histoarquitectura pulmonar y reducción de la hiperplasia de células cápsulas.

Conclusiones:

  • La terapia combinada SECU y HONK ofrece beneficios sinérgicos en el tratamiento del asma alérgica con ILA.
  • El enfoque combinatorio se dirige efectivamente a múltiples mediadores inflamatorios.
  • Se necesitan más investigaciones para optimizar los protocolos clínicos para las afecciones inflamatorias respiratorias graves.
