Los macrófagos SPP1+ promueven el cáncer de cabeza y cuello al liberar SPP1, que interactúa con CD44 en las células tumorales. Este eje SPP1-CD44 mejora el tallo del tumor y la migración, formando un bucle de retroalimentación positiva.

Sus antecedentes:
- Los macrófagos SPP1+ están implicados en la progresión del carcinoma de células escamosas de cabeza y cuello (HNSCC).
- Los mecanismos precisos por los que estos macrófagos impulsan el HNSCC siguen sin estar claros.
Objetivo del estudio:
- Para dilucidar la interacción entre los macrófagos SPP1+ y las células tumorales en HNSCC.
- Investigar el papel del eje SPP1-CD44 en la regulación de la estirpe y la metástasis del HNSCC.
Principales métodos:
- Se utilizaron inmunohistoquímica múltiple, co-inmunoprecipitación e inmunocitoquímica para detectar las interacciones SPP1-CD44.
- La citometría de flujo, la RT-PCR, la formación de esferoides, Transwell y los ensayos de cicatrización de heridas evaluaron la capacidad de las células madre tumorales y su migración.
- Western blot analizó la vía PI3K/Akt; ELISA midió el SPP1 sérico y el CD44 soluble.
Principales resultados:
- Se confirmaron las interacciones SPP1 y CD44 entre los macrófagos SPP1+ y las células HNSCC.
- La SPP1 liberada por los macrófagos promueve la metástasis de las células madre tumorales a través del eje SPP1-CD44.
- Existe un bucle de retroalimentación positiva donde el CD44 soluble regula la expresión del macrófago SPP1.
Conclusiones:
- La interacción SPP1-CD44 mejora significativamente la capacidad de las células madre tumorales y la migración en el HNSCC.
- Los macrófagos SPP1+ y las células tumorales participan en un ciclo de retroalimentación positiva mediado por SPP1-CD44, impulsando la progresión del HNSCC.
