Sus antecedentes:

  • La fisiología de los mamíferos está gobernada por un sistema de cronometraje circadiano jerárquico, con el núcleo supraquiasmático (SCN) como el marcapasos maestro.
  • El hígado, crucial para la adaptación metabólica, muestra ritmos diarios en la expresión génica, la síntesis de proteínas y la desintoxicación.
  • Sorprendentemente, la masa hepática y la morfología de los hepatocitos oscilan significativamente durante un período de 24 horas.

Objetivo del estudio:

  • Investigar los mecanismos subyacentes a la oscilación diurna de la masa hepática y el tamaño de los hepatocitos.
  • Explorar el papel del citoesqueleto de actina y las vías de señalización asociadas en la regulación de estos ritmos circadianos.
  • Para entender cómo estos cambios celulares contribuyen a la sincronización de los relojes del hígado.

Principales métodos:

  • Se utilizaron modelos de ratón para estudiar las fluctuaciones diarias de la masa hepática, el tamaño de los hepatocitos, el ARN y la acumulación de proteínas.
  • El contenido de ribosomas analizado y su correlación con las tasas de síntesis de proteínas.
  • Se investigó la dinámica del citoesqueleto de actina y su regulación por medio de la señalización de factores de transcripción-factor de respuesta sérica (MRTF-SRF) relacionados con la miocardina.

Principales resultados:

  • La masa hepática en ratones fluctúa entre un 30 y un 40% diariamente, acompañada de cambios rítmicos en el tamaño de los hepatocitos y en los niveles globales de proteínas y ARN.
  • La abundancia de ribosomas se correlaciona con el tamaño del hígado, actuando como un factor limitante de la velocidad de síntesis de proteínas diarias.
  • El citoesqueleto de actina de los hepatocitos sufre ciclos de polimerización significativos, con la formación de haces de F-actina durante la noche, coincidiendo con el tamaño máximo del hígado.

Conclusiones:

  • Los ritmos circadianos en la masa hepática son impulsados por oscilaciones en el tamaño de los hepatocitos y el contenido de ribosomas, lo que afecta a la síntesis de proteínas.
  • Los ciclos de polimerización de la actina en los hepatocitos son esenciales para mantener la robustez celular durante las fluctuaciones de tamaño.
  • La señalización MRTF-SRF, influenciada por la dinámica de la actina, juega un papel en la transcripción circadiana de genes de reloj como Per2, sincronizando los relojes hepáticos.
