Immunotherapy is a treatment that boosts or manipulates the immune system to fight diseases, including cancer. Sus antecedentes:

  • Las células T gamma delta (γδ) poseen mecanismos citotóxicos duales: citotoxicidad independiente de anticuerpos (AIC) y citotoxicidad celular dependiente de anticuerpos (ADCC).
  • Comprender la interacción entre la variabilidad del donante de células T, la heterogeneidad del tumor y la muerte de células T γδ es crucial para avanzar en las inmunoterapias.

Objetivo del estudio:

  • Investigar cómo la heterogeneidad entre donantes (IDH) de las células T γδ y la heterogeneidad entre tumores (ITH) del cáncer colorrectal (CRC) influyen en los mecanismos citotóxicos multimodales.
  • Evaluar la eficacia de las células T γδ de ingeniería en la superación de la evasión inmune mediada por células cancerosas.

Principales métodos:

  • Análisis sistemático de fenoescape de una sola célula de más de 1.000 coculturas de células T γδ y organoides de cáncer colorrectal derivados de pacientes.
  • Análisis de la señalización de la modificación post-traducional (PTM), el estado del ciclo celular, la apoptosis y los inmunofenotipos de las células T.
  • Se utilizaron células T γδ de la proteína de fusión IL-15Rα-IL-15 de ingeniería y se evaluó el ADCC dirigido a B7-H3.

Principales resultados:

  • Las células T γδ stIL15 modificadas mostraron AIC frente a las DOP sin apoyo de citoquinas externas.
  • Las células de cáncer colorrectal se adaptaron al AIC mediante la recableación de las redes de señalización PTM de las células γδ T de una manera dependiente de ITH, suprimiendo la citotoxicidad.
  • La combinación de AIC con ADCC dirigido a B7-H3 permitió a las células stIL15 γδ T superar la inmunomodulación inducida por el cáncer y matar las células madre del cáncer de colon quimiorresistentes.

Conclusiones:

  • La citotoxicidad multimodal, que combina AIC y ADCC, es esencial para que las células γδ T superen las estrategias de evasión inmunológica específicas de ITH empleadas por las células cancerosas.
  • Las células γδ T diseñadas con mecanismos citotóxicos combinados demuestran potencial para atacar las células madre cancerosas quimiorresistentes, allanando el camino para nuevos enfoques inmunoterapéuticos.
