Sus antecedentes:

  • La señalización de AKT mediada por fosfoinositida (PIP) regula los procesos celulares fundamentales como el metabolismo, la supervivencia, la proliferación y la motilidad.
  • La desregulación de la señalización de AKT está implicada en varias patologías, especialmente el cáncer.
  • Si bien la activación de AKT citosólico está bien estudiada, sus funciones nucleares son un área emergente de investigación.

Objetivo del estudio:

  • Revisar la regulación espacial de la señalización de AKT por los fosfoinosítidos (PIP) en diferentes compartimentos celulares.
  • Aclarar los mecanismos de la compartimentación de la TCA y sus consecuencias funcionales.
  • Resaltar el papel de la regulación espacial de la TCA impulsada por el PIP en la oncogénesis y la quimiorresistencia.

Principales métodos:

  • Revisión de la literatura centrada en la señalización de AKT mediada por fosfoinosítidos.
  • Análisis de la localización y la función de AKT en los microdominios de la membrana plasmática, los endosomas y el núcleo.
  • Exploración de andamios de proteínas y de los señalosomas para refinar la especificidad de la señal AKT.

Principales resultados:

  • La activación de AKT ocurre en microdominios especializados y en las uniones de células en la membrana plasmática, lo que influye en la polaridad celular, la adhesión y la migración.
  • En los endosomas, los PIP integran la señalización AKT con el tráfico intracelular y la organización citoesquelética.
  • El AKT nuclear interactúa con el señalosoma y las quinasas p53-PIP, regulando la oncogénesis y la quimioresistencia.

Conclusiones:

  • La compartimentación espacial impulsada por PIP de AKT es un determinante crítico de las respuestas celulares y la oncogénesis.
  • Comprender la localización y regulación de AKT dentro de diferentes compartimentos celulares ofrece información sobre la biología del cáncer.
  • Elucidar estos mecanismos puede revelar nuevas estrategias terapéuticas para el tratamiento del cáncer.
