Sus antecedentes:
- Las enfermedades cerebrovasculares (CEV) y las enfermedades neurodegenerativas (ND) son una carga significativa para la salud mundial.
- Existe una relación compleja entre CeVD y ND, pero sus fundamentos genéticos compartidos no se comprenden bien.
Objetivo del estudio:
- Investigar la arquitectura genética compartida y la pleiotropía entre las enfermedades neurodegenerativas y las enfermedades cerebrovasculares, incluido el accidente cerebrovascular.
- Identificar los factores genéticos compartidos y los mecanismos biológicos que contribuyen tanto a la ECV como a la EN.
Principales métodos:
- Se realizaron análisis de correlación genética y pleiotropía en todo el genoma.
- Se utilizaron estadísticas resumidas de estudios de asociación de todo el genoma a gran escala (GWAS) de poblaciones de ascendencia cruzada, europeas y del sur de Asia (SAS).
- Para el análisis se utilizaron las herramientas estadísticas LDSC, PLACO y COLOC.
Principales resultados:
- Se identificaron 116 loci genéticos compartidos y 40 regiones genéticas causales compartidas entre múltiples pares de CeVD y ND.
- Se detectaron regiones genéticas altamente pleiotrópicas cerca de los genes ICA1L, HLA-DQA1, HLA-DRB1, MIR297 y PITX2.
- ERGIC1 fue identificado como un nuevo locus causal compartido entre la esclerosis lateral amiotrófica y el accidente cerebrovascular de los vasos pequeños.
Conclusiones:
- Esta investigación descubre nuevos loci genéticos compartidos y vías entre ND y CeVD.
- Los hallazgos contribuyen a una comprensión más profunda de los mecanismos biológicos subyacentes en estos trastornos neurológicos.
- El estudio pone de relieve el potencial para identificar objetivos terapéuticos comunes para ambas categorías de enfermedades.
