Sus antecedentes:

  • La miocarditis viral, una inflamación del corazón por infecciones virales, tiene malos resultados debido a los tratamientos limitados.
  • La muerte de los cardiomiocitos es un sello distintivo, pero sus mecanismos y objetivos terapéuticos no están claros.
  • La medicina tradicional china (MTC) carece de tratamientos aprobados para la miocarditis viral.

Objetivo del estudio:

  • Investigar el potencial terapéutico de las inyecciones de patente de MTC para la miocarditis viral.
  • Identificar los mecanismos subyacentes a los efectos protectores de Danhong Injection (DHI).
  • Evaluar la eficacia del DHI en la reducción de la lesión cardíaca inducida por la miocarditis viral.

Principales métodos:

  • Se examinó la eficacia de seis inyecciones de patente de MTC contra la muerte de los cardiomiocitos inducida por el virus Coxsackie B3 (CVB3).
  • Se administró DHI a un modelo de ratón infectado por CVB3 con miocarditis viral.
  • Se investigó el mecanismo de la DHI mediante la evaluación de la inhibición y fosforilación de la proteína quinasa II Ca2+/calmodulina-dependiente (CaMKII) en muestras de pacientes y células humanas.

Principales resultados:

  • Danhong Injection (DHI) redujo significativamente la muerte de los cardiomiocitos in vitro e in vivo.
  • El DHI alivió la inflamación, la disfunción cardíaca y la mortalidad en un modelo de ratón con miocarditis viral.
  • DHI inhibió la fosforilación de CaMKII, un factor clave en la muerte de los cardiomiocitos, y demostró efectos antiinflamatorios.

Conclusiones:

  • La muerte de los cardiomiocitos mediada por CaMKII es fundamental para la progresión de la miocarditis viral.
  • Danhong Injection (DHI) es un agente terapéutico prometedor para la miocarditis viral.
  • La eficacia de DHI se deriva de la inhibición de CaMKII y las propiedades antiinflamatorias, ofreciendo una nueva estrategia de tratamiento.
