Sus antecedentes:

  • Las terapias de ARN mensajero (ARNm) son prometedoras para las terapias de reemplazo de genes y proteínas.
  • Los métodos actuales para la administración de ARNm utilizando nanopartículas lipídicas (LNP) son efectivos para el hígado, pero limitados para otros órganos.
  • La administración eficiente de ARNm extrahepático por órganos sigue siendo un desafío crítico para aplicaciones terapéuticas más amplias.

Objetivo del estudio:

  • Desarrollar un método novedoso para lograr la entrega de ARNm selectivo de órganos a órganos extrahepáticos después de la administración sistémica.
  • Diseñar nanopartículas lipídicas (LNP) con capacidades de orientación mejoradas para tejidos no hepáticos.
  • Establecer una plataforma versátil para dirigir el tropismo de ARNm a órganos específicos más allá del hígado.

Principales métodos:

  • Se introdujo un método de orientación selectiva de órganos codificado por péptidos (POST) que implica el ajuste modular de LNP a través de la ingeniería de superficie con secuencias específicas de aminoácidos (códigos POST).
  • Se utilizaron simulaciones de dinámica molecular para optimizar las afinidades de unión péptido-proteína para la formación de corona de proteínas plasmáticas específicas.
  • Pruebas in vitro e in vivo realizadas para validar la eficacia de los LNP modificados por POST en la selección de órganos.

Principales resultados:

  • Se ha demostrado que POST permite la administración selectiva de ARNm a órganos extrahepáticos después de la administración sistémica.
  • Se demostró que la especificidad de la orientación surge de la corona proteica única formada en LNP decorados con péptidos, influenciada por la secuencia de péptidos y las interacciones de proteínas plasmáticas.
  • Confirmó la aplicabilidad de la plataforma POST para la entrega de varios ácidos ribonucleicos y herramientas de edición de genes.

Conclusiones:

  • La plataforma POST proporciona un enfoque modular para la ingeniería de superficie LNP para lograr un tropismo de órganos preciso.
  • Este método amplía significativamente el alcance y la versatilidad de la administración de ARNm selectivo de órganos para aplicaciones terapéuticas.
  • La tecnología POST representa un avance clave para superar las limitaciones de la administración de ARNm extrahepático.
