Sus antecedentes:

  • Las modificaciones epigenéticas, incluida la metilación del ADN, son reguladores cruciales de las respuestas inmunes.

  • La enfermedad de Graves (GD) es un trastorno autoinmune que afecta a la glándula tiroides.

Objetivo del estudio:

  • Para investigar el panorama de la metilación del ADN del gen IL10 en pacientes con enfermedad de Graves.
  • Explorar el potencial de los patrones de metilación de la IL10 como biomarcadores para el diagnóstico y el monitoreo de la DG.

Principales métodos:

  • Perfiles cuantitativos de metilación del ADN del gen IL10 en muestras de sangre periférica de pacientes con GD y controles sanos.
  • Construcción de modelos de aprendizaje automático utilizando características de metilación de CpG para la clasificación de enfermedades.
  • Análisis de correlación entre la metilación específica del sitio de CpG y los indicadores clínicos de GD.

Principales resultados:

  • Se observó una metilación del gen IL10 significativamente elevada en pacientes con DM en comparación con los controles sanos.
  • Se identificó un grupo hipermetilado distinto dentro del gen IL10 en GD.
  • La metilación específica del sitio se correlacionó positivamente con los niveles de hormona tiroidea (FT3) y los anticuerpos del receptor de hormona estimulante de la tiroides (TRAb), lo que indica una asociación con la actividad de la enfermedad.

Conclusiones:

  • Distintos patrones de metilación del gen IL10 están asociados con la enfermedad de Graves.
  • Los sitios específicos de CpG dentro del gen IL10 pueden servir como biomarcadores potenciales para el diagnóstico, la estratificación y el monitoreo de GD.
  • Comprender la metilación de IL10 en GD ofrece información sobre la patogénesis de la enfermedad y la regulación inmune.
