Sus antecedentes:

  • Se sabe que los lisosomas importan cisteína, pero su importancia fisiológica no está clara.
  • MFSD12 es una proteína transmembrana esencial para la importación de cisteína en los lisosomas y los melanosomas.

Objetivo del estudio:

  • Investigar el papel fisiológico de la importación de cisteína lisosomal mediada por MFSD12 en el desarrollo de todo el organismo.
  • Para aclarar la función específica de la importación de cisteína dentro del lisosoma.

Principales métodos:

  • Generación y análisis de ratones knockout con MFSD12.
  • Análisis de la expresión génica para identificar las vías celulares afectadas.
  • Tratamiento materno con cisteamina para evaluar los efectos de rescate.

Principales resultados:

  • Los ratones knockout MFSD12 muestran letalidad embrionaria debido a defectos de organogénesis.
  • La pérdida de MFSD12 conduce a una expresión alterada de genes relacionados con el estrés celular y el metabolismo del tiol.
