Objetivo del estudio:
- Identificar los genes asociados con la palmitoilación que están implicados en el TAG.
- Investigar el papel mecanicista de estos genes, centrándose particularmente en las vías mediadas por las células inmunes.
Principales métodos:
- Identificación de genes de loci de rasgos cuantitativos de expresión de la palmitoilación (eQTL) mediante la intersección de genes relacionados con la palmitoilación con conjuntos de datos de eQTL de todo el genoma.
- Se empleó la aleatorización mendeliana de dos muestras (MR) y la aleatorización mendeliana basada en datos resumidos (SMR) para establecer relaciones causales entre los genes eQTL y GAD.
- Se examinaron los mecanismos subyacentes a través de procesos mediados por células inmunes.
Principales resultados:
- Se identificaron 22 loci del gen de la palmitoilación-eQTL, de los cuales cinco están significativamente relacionados con la GAD: PPT2, ZDHHC5, ZDHHC13, ZDHHC20 y ZDHHC24.
- ZDHHC5 y ZDHHC13 fueron validados como genes diana mediante análisis de SMR.
- El análisis de RM de mediación reveló que SSC-A en las células T CD4+ medió el efecto de ZDHHC5 (mediación del 36,4%).
Conclusiones:
- ZDHHC5 está implicado en la patogénesis de la GAD.
- ZDHHC5 modula la SSC-A en las células T CD4+, lo que sugiere un nuevo mecanismo mediado por células inmunes en la GAD.
- Estos hallazgos ofrecen nuevos objetivos para la investigación de la fisiopatología del TAG.
