Sus antecedentes:

  • Los genomas de los mamíferos suelen estar metilados por ADN, pero los ovocitos mantienen bajos niveles de metilación del ADN (DNAme), principalmente en regiones transcritas.
  • Los mecanismos que controlan el ADN de novo en los ovocitos y su impacto en el desarrollo embrionario siguen siendo en gran medida desconocidos.

Objetivo del estudio:

  • Para investigar los mecanismos que restringen el ADN de novo en los ovocitos.
  • Determinar el papel de KDM2A y KDM2B en la regulación del metiloma de los ovocitos.
  • Evaluar las consecuencias del ADN aberrante de los ovocitos en el desarrollo embrionario.

Principales métodos:

  • Utilizó mutaciones genéticas en KDM2A y KDM2B en ratones.
  • Analizó patrones de metilación del ADN en ovocitos y embriones tempranos.
  • Evaluación de la transcripción génica y el desarrollo embrionario en embriones mutantes.

Principales resultados:

  • KDM2A y KDM2B impiden la di-metilación de la histona H3 lisina 36 en todo el genoma, que normalmente impide el ADNme catalizado por DNMT3A.
  • La pérdida de KDM2A/KDM2B conduce a un ADNme aberrante en las islas CpG en los ovocitos.
  • Este ADN materno aberrante reprime la transcripción génica en embriones de dos células y perjudica el desarrollo pre-implantación.
  • La deficiencia de Dnmt3a durante la oogénesis rescata los defectos de desarrollo causados por el ADN materno aberrante.

Conclusiones:

  • KDM2A y KDM2B son esenciales para mantener el metiloma de los ovocitos y garantizar la competencia del desarrollo.
  • La capacidad de reprogramación de los primeros embriones es insuficiente para borrar el ADN materno aberrante.
  • El desarrollo embrionario temprano es sensible a los efectos de dosificación genética que surgen de la desregulación epigenética.
