Resumen

Los agentes de contraste basados en gadolinio (GBCA) muestran una unión cooperativa variable a la proteína lisozima. El análisis frontal con electroforesis capilar continua (FACCE, por sus siglas en inglés) analiza eficazmente estas interacciones, revelando la fuerza de unión y la estequiometría para mejorar la seguridad del agente de contraste de RM.

Sus antecedentes:

  • Los agentes de contraste basados en gadolinio (GBCA) son cruciales para la resonancia magnética (MRI).
  • Comprender las interacciones de GBCA con macromoléculas biológicas es vital para la seguridad y el diseño de agentes de próxima generación.
  • Se necesitan métodos analíticos sólidos para estudiar estas interacciones.

Objetivo del estudio:

  • Investigar las interacciones de unión entre lisozima y varios GBCA utilizando el análisis frontal con electroforesis capilar continua (FACCE).
  • Para determinar la estequiometría, la fuerza de unión y la cooperatividad de estas interacciones.
  • Evaluar la idoneidad de FACCE para el análisis de las interacciones GBCA-proteína.

Principales métodos:

  • Se empleó la electroforesis capilar continua de análisis frontal (FACCE).
  • La lisozima fue utilizada como proteína modelo.
  • Se analizaron varios GBCA: Gd-DTPA, Gd-BOPTA, Gd-DOTA, Gd-PCTA D2 y Gd-HP-DO3A.

Principales resultados:

  • Los GBCA exhibieron diferentes grados de unión cooperativa a la lisozima.
  • Se observó estequiometría (n=4) para Gd-DTPA, Gd-BOPTA y Gd-DOTA.
  • La fuerza de unión siguió el orden: Gd-BOPTA > Gd-DTPA > Gd-DOTA. Gd-PCTA D2 no mostró ninguna unión significativa.
  • FACCE no pudo resolver las interacciones para Gd-HP-DO3A debido a la interferencia.

Conclusiones:

  • FACCE es un método adecuado para estudiar las interacciones GBCA-proteína, que requiere un volumen de muestra mínimo.
  • El método permite la determinación de la estequiometría, las constantes de unión y la cooperatividad.
  • Las interacciones GBCA-lizozima son cooperativas, y el modelo de Hill las analiza efectivamente dentro del rango de concentración probado.
