These disturbances can lead to various symptoms, depending on their severity and the underlying cause. Some common factors contributing to arrhythmias include hypoxia, ischemia, electrolyte imbalances, excessive catecholamine exposure, drug toxicity, and muscle overstretching. Resumen

Los neutrófilos promueven arritmias cardíacas como la taquicardia ventricular a través de la molécula gamma parecida a la resistin (RELMγ). La eliminación de RELMγ en ratones redujo estas peligrosas alteraciones del ritmo cardíaco, revelando un nuevo mecanismo para la muerte súbita cardíaca.

Sus antecedentes:

  • La taquicardia ventricular (VT) es una arritmia potencialmente mortal asociada con el infarto de miocardio.
  • Los neutrófilos se infiltran en el corazón isquémico y contribuyen al desarrollo de VT.
  • Los mecanismos moleculares específicos por los cuales los neutrófilos promueven la VT siguen siendo incompletamente entendidos.

Objetivo del estudio:

  • Investigar el papel de los genes expresados diferencialmente en los neutrófilos durante la TE inducida por infarto de miocardio.
  • Identificar nuevos objetivos moleculares para la prevención o el tratamiento de la VT.

Principales métodos:

  • Análisis comparativo de la expresión génica de neutrófilos y macrófagos en modelos de infarto de ratón.
  • La generación de ratones retnlg knockout específicos de los leucocitos.
  • Evaluación de la electrofisiología cardíaca y de la integridad cardiomiocitaria en ratones nocaut y de tipo salvaje.
  • Estudios in vitro con liposomas y células de mamíferos para evaluar los efectos disruptores de la membrana de la molécula gamma parecida a la resistina (RELMγ) y la resistina humana.

Principales resultados:

  • La molécula gamma parecida a la resistina (RELMγ) se regulaba significativamente en los neutrófilos dentro de la zona del infarto.
  • La deleción específica de leucocitos de *Retnlg* redujo notablemente la incidencia de VT en ratones con infarto agudo de miocardio.
  • RELMγ indujo la permeabilidad de la membrana en los cardiomiocitos, lo que condujo a un retraso en la despolarización y posterior muerte celular, ambos desencadenantes aritmogénicos potentes.
  • La resistina humana demostró una actividad disruptiva de membrana similar en los liposomas y las células de mamíferos.

Conclusiones:

  • RELMγ, una proteína formadora de poros, es un mediador clave del VT inducido por neutrófilos después del infarto de miocardio.
  • El mecanismo implica daño directo a las membranas de los cardiomiocitos, creando inestabilidad eléctrica.
  • La orientación de RELMγ representa una estrategia terapéutica potencial para prevenir la muerte súbita cardíaca asociada con arritmias.
