Resumen
La proteína MCL1 no parece aumentar la quimiorresistencia o moverse hacia el núcleo durante el tratamiento con fármacos contra el cáncer, al contrario de lo que se pensaba anteriormente. Se necesitan más estudios para comprender las funciones nucleares de MCL1.

Sus antecedentes:
- El miembro anti-apoptótico de la familia BCL2 MCL1 es frecuentemente sobre expresado en cánceres y asociado con resistencia al tratamiento.
- MCL1 tiene funciones mitocondriales conocidas y roles nucleares propuestos en la quimiorresistencia, el ciclo celular y la reparación del ADN.
Objetivo del estudio:
- Investigar las funciones nucleares propuestas de MCL1 en la quimiorresistencia.
- Para determinar si MCL1 se transloca al núcleo e influye en la quimiorresistencia durante la quimioterapia.
Principales métodos:
- Utilizó un anticuerpo monoclonal recientemente validado contra MCL1 en varias líneas celulares de cáncer.
- Se utiliza la biotinilación de proximidad para identificar los interactores nucleares MCL1.
- Se evaluó la localización de MCL1 y su impacto en la quimioresistencia después de la exposición al fármaco.
Principales resultados:
- No se encontró evidencia de que MCL1 mejore la quimiorresistencia o se acumule en el núcleo después del tratamiento farmacológico.
- Se identificaron nuevos interactores nucleares MCL1, pero no se recuperaron los socios de reparación del ADN y el ciclo celular informados anteriormente.
- Los datos no apoyan el papel de MCL1 en la conferencia de quimiorresistencia a través de mecanismos nucleares.
Conclusiones:
- El estudio desafía los hallazgos anteriores que implican a la MCL1 nuclear en la quimiorresistencia.
- No se observó la translocación nuclear de MCL1 ni la interacción con las proteínas reparadoras del ADN durante la quimioterapia.
- Se requiere más investigación para aclarar el significado funcional de la MCL1 nuclear.
