Sus antecedentes:

  • La migración celular a lo largo de gradientes de rigidez (durotaxis) es crucial para el desarrollo y la metástasis del cáncer.
  • Los mecanismos precisos que rigen la durotaxis y su universalidad en todos los tipos de células siguen siendo incompletamente entendidos.

Objetivo del estudio:

  • Investigar las funciones de las fuerzas de tracción celular y la persistencia direccional en la migración celular a lo largo de gradientes de rigidez.
  • Para dilucidar un mecanismo unificado para el sesgo durotaxis a través de diferentes tipos de células y comportamientos de migración.

Principales métodos:

  • Se utilizan hidrogeles sintonizables de polietileno glicol (PEG) con gradientes de rigidez definidos (1-16 kPa).
  • Perturbación de la contractilidad celular (mediante el uso de blebbistatina, oligomicina) y de la adhesión (mediante el uso de mutantes de vinculina) en las células cancerosas.
  • Desarrolló y validó un modelo computacional que integra las fuerzas de tracción y la estabilidad de la F-actina para predecir la durotaxis.

Principales resultados:

  • Las células con altas fuerzas de tracción exhibieron una migración persistente hacia entornos más rígidos (durotaxis positiva).
  • Las células con fuerzas de tracción reducidas perdieron persistencia y migraron hacia entornos más suaves (durotaxis negativa).
  • El modelo computacional vinculó con éxito la tracción dependiente de la rigidez a la persistencia, capturando ambos comportamientos de durotaxis con un solo conjunto de parámetros.

Conclusiones:

  • Se estableció un mecanismo unificado en el que la persistencia regulada por la tracción rige el sesgo de la durotaxis, aplicable a todos los tipos de células.
  • Se ha demostrado que la modulación de la actividad de la miosina o el refuerzo de la adhesión pueden cambiar los estados de durotaxis.
  • Los hallazgos proporcionan información para el diseño de biomateriales para la migración celular dirigida y el desarrollo de estrategias terapéuticas para la reparación de tejidos y el cáncer.
