Resumen
El potenciador del zeste homólogo 2 (EZH2) regula los ARN largos no codificantes clave (ARNlnc) en el cáncer gástrico (GC). Dirigirse al eje EZH2/lncRNA ofrece una estrategia terapéutica potencial para la progresión de la GC.

Sus antecedentes:
- El cáncer gástrico (GC) sigue siendo una de las principales causas de mortalidad por cáncer en todo el mundo.
- El potenciador del zeste homólogo 2 (EZH2) es un regulador oncogénico clave sobreexpresado en muchos cánceres.
- Los ARN largos no codificantes (ARNlnc) son críticos en la regulación génica y están implicados en el desarrollo del cáncer.
Objetivo del estudio:
- Para investigar los lncRNA asociados a EZH2 en el cáncer gástrico.
- Explorar el papel regulador de estos lncRNA en la progresión de la GC.
- Identificar posibles objetivos terapéuticos dentro de la red EZH2/lncRNA.
Principales métodos:
- Se utilizaron datos de TCGA RNA-seq y miRNA-seq para la expresión diferencial y los análisis de correlación.
- Construyó una red de ARN endógeno (ceRNA) competitivo de lncRNAs, miRNAs y mRNAs.
- Se han realizado experimentos de genómica funcional, incluida la eliminación/sobreexpresión de EZH2 en líneas celulares GC y la validación por RT-qPCR.
Principales resultados:
- Se identificaron 16 lncRNAs regulados hacia arriba y 8 regulados hacia abajo significativamente correlacionados con la expresión de EZH2.
- El EZH2 confirmado modula la expresión de lncRNAs específicos (por ejemplo, PVT1, MNX1-AS1, LINC01235) en las células GC.
- Se demostró la influencia reguladora de EZH2 en una red de lncRNAs crucial para el cáncer gástrico.

- El eje EZH2/lncRNA juega un papel importante en la progresión del cáncer gástrico.
