Sus antecedentes:

  • La diversidad de las isoformas de ARN mensajero (ARNm) surge de la iniciación y terminación de la transcripción.
  • La interacción entre los sitios de inicio de transcripción (TSS) y los sitios finales de transcripción (TES) no se comprende bien.

Objetivo del estudio:

  • Investigar la relación entre el inicio de la transcripción y el uso del sitio final.
  • Aclarar los mecanismos que rigen la diversidad de las isoformas de ARNm.

Principales métodos:

  • Perfilamiento sistemático del uso conjunto de los sitios de inicio y finalización de la transcripción.
  • Análisis de la longitud del gen, las características de la cromatina y la velocidad de tráfico de ARN polimerasa II.

Principales resultados:

  • El mRNA que utiliza TSS aguas arriba utiliza preferentemente TES aguas arriba, y los sitios aguas abajo están acoplados de manera similar.
  • Un eje de iniciación y terminación posicional (PITA, por sus siglas en inglés) describe el uso alternativo acoplado de TES basado en el orden genómico.
  • El PITA prevalece en genes más largos con características específicas de la cromatina.
  • La selección del sitio inicial del ARNm 5' afecta a la selección del sitio final del 3', influenciada por la velocidad de la ARN polimerasa II.

Conclusiones:

  • La organización espacial y la dinámica de transcripción vinculan la iniciación de la transcripción y las decisiones del extremo 3' del ARNm.
  • Estos eventos acoplados definen los patrones de expresión de las isoformas de ARNm.
  • El modelo PITA proporciona un marco para comprender la selección coordinada de TSS y TES.
