Sus antecedentes:

  • El microambiente inmune del tumor (TIME) influye en la progresión del cáncer y los resultados del tratamiento.
  • La inmunofluorescencia múltiple (mIF) es valiosa para el análisis TIME, pero es costosa y de bajo rendimiento.
  • Los métodos existentes luchan con la caracterización TIME a gran escala debido a las limitaciones de datos.

Objetivo del estudio:

  • Desarrollar GigaTIME, un marco de IA para el modelado de tiempo a escala de población.
  • Para unir la morfología celular y los estados usando un traductor trans-modal.
  • Para generar imágenes mIF virtuales de diapositivas de H&E para un análisis extenso.

Principales métodos:

  • Entrenado un traductor trans-modal en 40 millones de células con datos de H&E y mIF emparejados (21 proteínas).
  • Aplicado GigaTIME a 14.256 pacientes en 24 tipos de cáncer, generando 299.376 diapositivas mIF virtuales.
  • Los hallazgos validados en 10.200 pacientes con TCGA.

Principales resultados:

  • Generó una gran cohorte virtual de datos de tiempo.
  • Se descubrieron 1.234 asociaciones significativas entre proteínas, biomarcadores, estadificación y supervivencia.
  • Demostró la viabilidad de un análisis de tiempo a gran escala, previamente limitado por la escasez de datos de MIF.

Conclusiones:

  • GigaTIME permite un análisis de tiempo escalable y rentable utilizando diapositivas H&E estándar.
  • El marco de IA expande significativamente el alcance de la investigación TIME.
  • Este enfoque facilita el descubrimiento de nuevos biomarcadores y objetivos terapéuticos.
