  • Comparar la magnitud y distribución de la señal meníngea de [F-18]MK6240 entre portadores y no portadores de APOE4.; Investigar la influencia del portador de APOE4 en la imagenología PET tau.; Evaluar posibles sesgos en la cuantificación de la PET de la enfermedad de Alzheimer.

Principales métodos:

  • Se escanearon 1051 participantes mediante PET en escáneres Biograph Horizon mCT o ECAT HR+.; Se generaron imágenes SUVR utilizando una canalización estandarizada, con una máscara cortical diluida de 5 mm utilizada para el análisis de ROI de las meninges.; La regresión múltiple comparó el SUVR meníngeo entre portadores y no portadores de APOE4, incluyendo el sexo y el modelo del escáner como covariables.

Principales resultados:

  • Se observó una mayor señal meníngea promedio de [F-18]MK6240 en portadores de APOE4 (p=0,03), mujeres (p<0,001) y en el escáner mCT (p<0,001).; No se encontraron términos de interacción significativos entre las variables.; El análisis de sensibilidad en mujeres reveló una señal meníngea significativamente mayor en portadores de APOE4 en el HR+ (p=0,009) y mCT (p=0,05).

Conclusiones:

  • La variabilidad de la señal meníngea de [F-18]MK6240 puede sesgar los resultados de la PET tau en estudios de enfermedad de Alzheimer.; El portador de APOE4 y el sexo son factores significativos que influyen en la señal meníngea.; Estas relaciones deben considerarse para una cuantificación precisa de MK6240 y comparaciones poblacionales en la investigación de la EA.
