Objetivo del estudio:

  • Examinar las asociaciones entre los biomarcadores plasmáticos cuantificados por NULISA™ y el rendimiento cognitivo.; Investigar el valor predictivo de estos biomarcadores para la demencia incidente en adultos mayores.

Principales métodos:

  • Se utilizaron datos de 116 participantes de la cohorte ARIC.; Se cuantificaron biomarcadores plasmáticos utilizando la plataforma de proteínas NULISA™ CNS 126, que incluye Aβ, p-tau, NfL y GFAP.; Se evaluó el rendimiento cognitivo y se identificaron casos de demencia incidente, utilizando modelos de regresión ajustados por covariables.

Principales resultados:

  • El factor de crecimiento y diferenciación 15 fue la única proteína asociada con la demencia incidente (β = 4,7, p < 0,001).; El antígeno CD63 (β = -0,43, p < 0,001) y la Eotaxina CCL11 (β = -0,37, p < 0,001) se asociaron con la cognición global.; La proteína alfa regulada por crecimiento CXCL1 se asoció con la función ejecutiva (β = -0,33, p < 0,001).

Conclusiones:

  • Las proteínas involucradas en la exocitosis de vesículas sinápticas y la inflamación están relacionadas con la cognición y el riesgo de demencia.; El poder del estudio fue limitado, lo que requiere tamaños de muestra más grandes para la identificación de vías proteómicas más amplias.; La investigación futura debe centrarse en cohortes más grandes para descubrir nuevas vías proteómicas relacionadas con el deterioro cognitivo y la demencia.
