Principales métodos:

  • Se utilizaron datos de 386 participantes del consorcio ADNI FNIH durante 11 años.
  • Se analizaron las trayectorias longitudinales de 5 ensayos de p-tau217 y 4 ensayos de Aβ42/40 con múltiples escaneos de Aβ-PET.
  • Se emplearon modelos de riesgos proporcionales de Cox para vincular los datos de biomarcadores basales y longitudinales con la progresión de la positividad de Aβ-PET.

Principales resultados:

  • Los ensayos de p-tau217 en plasma, a diferencia de los ensayos de Aβ42/40, mostraron aumentos a lo largo del tiempo tanto en el grupo Aβ-PET negativo como en el positivo.
  • La mayor tasa de cambio en p-tau217 en plasma se asoció con un mayor riesgo de progresión a la positividad de Aβ-PET en individuos Aβ-negativos.
  • La tasa de cambio en p-tau217 fue un predictor más fuerte de la positividad futura de Aβ-PET que los niveles basales.

Conclusiones:

  • Los niveles de p-tau217 en plasma demuestran cambios longitudinales, mientras que los ensayos de Aβ42/40 no.
  • La tasa de cambio en p-tau217 en plasma es el predictor más potente de la positividad futura de Aβ-PET entre los individuos inicialmente Aβ-negativos.
