  • Explorar cómo los factores genéticos (amiloide, tau, genotipo ApoE) interactúan con la edad y el sexo en la función ejecutiva utilizando la Tarea Continua de Rendimiento (CPT).
  • Investigar la sensibilidad de las pruebas cognitivas de pantalla táctil a la disminución temprana relacionada con la EA en comparación con las pruebas neuropsicológicas estándar.

Principales métodos:

  • Utilización de pruebas homólogas de pantalla táctil, incluida la CPT, en modelos de ratón de EA de próxima generación y en humanos en riesgo de EA.
  • Empleo de modelos de ratón con genes de inserción humanizados (amiloide, tau, ApoE ε3/ε4) para estudiar interacciones genéticas con la edad y el sexo.
  • Integración de la CPT en el estudio longitudinal PREVENT-AD, recopilando datos de más de 425 personas con cognición intacta y antecedentes familiares de EA.

Principales resultados:

  • La CPT se ha integrado en el estudio PREVENT-AD, con datos adquiridos de más de 220 personas.
  • Se han recopilado datos longitudinales de CPT de aproximadamente 80 participantes en la cohorte PREVENT-AD.

Conclusiones:

  • Las pruebas cognitivas basadas en pantalla táctil en humanos y ratones dilucidan los mecanismos de riesgo de la EA.
  • Los datos de CPT en humanos y ratones de PREVENT-AD pueden revelar interacciones entre el genotipo ApoE, la edad y el sexo en la disfunción cognitiva temprana.
  • La expansión de las pruebas de pantalla táctil en consorcios a gran escala vinculará el riesgo genético, el rendimiento cognitivo y los marcadores neurobiológicos para avanzar en la comprensión y las intervenciones de la EA.
