Objetivo del estudio:

  • Investigar si la atrofia relacionada con tau afecta preferentemente a los surcos cerebrales y sus profundidades.
  • Examinar la relación entre la acumulación de tau, el adelgazamiento cortical y la función cognitiva en la EA y el MCI.
  • Explorar el papel de la conectividad hipocampal en la vulnerabilidad relacionada con tau.

Principales métodos:

  • Se utilizaron escáneres de RM de 3T y tau-PET de la cohorte de la Iniciativa de Neuroimagen de la Enfermedad de Alzheimer (ADNI).
  • Se correlacionó la SUVR de tau neocortical con el grosor cortical, analizando la superposición con giros, surcos y profundidades de surcos.
  • Se examinó la asociación entre los clústeres de atrofia relacionados con tau y el rendimiento de la memoria, junto con la cuantificación post mortem de tau y el análisis de conectividad en estado de reposo.

Principales resultados:

  • El adelgazamiento cortical asociado a tau se superpuso significativamente con los surcos, especialmente con las profundidades de los surcos (p<0.0001).
  • Las regiones de los surcos mostraron valores t relacionados con tau más altos en comparación con las regiones de los giros (p<0.001).
  • El volumen de los clústeres afectados por tau predijo fuertemente el rendimiento de la memoria (p=0.02) y mostró una mayor conectividad hipocampal.

Conclusiones:

  • El adelgazamiento cortical relacionado con tau ocurre predominantemente en los surcos, particularmente en sus profundidades.
  • Esta vulnerabilidad de los surcos es potencialmente impulsada por una mayor acumulación de tau y una mayor conectividad hipocampal dentro de estas regiones.
  • Los hallazgos resaltan los surcos como áreas críticas afectadas por la patología tau en la enfermedad de Alzheimer.
