Objetivo del estudio:

  • Aplicar un novedoso modelo de predicción de la edad cerebral para estimar la Brecha de Edad Cerebral (BAG) en muestras de la Iniciativa de Neuroimagen de la Enfermedad de Alzheimer (ADNI).
  • Investigar posibles diferencias sexuales en la BAG entre individuos amiloides positivos en varias categorías de diagnóstico.
  • Explorar la relación entre la BAG, el estado amiloide, las puntuaciones cognitivas y el sexo.

Principales métodos:

  • Se desarrolló un modelo de conjunto utilizando datos del Biobanco del Reino Unido (UKBB) y del Estudio de Envejecimiento de Mayo Clinic (MCSA) para la predicción de la edad cerebral a partir de RM T1w.
  • El modelo empleó preprocesamiento robusto, aumento de datos y regularización para la generalización.
  • El modelo entrenado se aplicó a muestras de ADNI para estimar la BAG sin ajuste fino.

Principales resultados:

  • No se observó una diferencia sexual significativa en la BAG en sujetos cognitivamente normales, lo que indica neutralidad del modelo.
  • La BAG aumentó a lo largo de la trayectoria de la enfermedad de Alzheimer (EA), de acuerdo con investigaciones previas.
  • Después de ajustar por edad y puntuaciones de ADAS13, las mujeres amiloides positivas mostraron cerebros significativamente de aspecto más viejo que los hombres en la mayoría de los grupos de diagnóstico (p < 0,05).

Conclusiones:

  • Los cerebros de mujeres parecen ser más viejos que los de hombres en la mayoría de los grupos de diagnóstico al considerar la positividad amiloide y las puntuaciones cognitivas.
  • Este hallazgo sugiere una diferencia potencial en la reserva cognitiva entre sexos, con mujeres que potencialmente tienen una mayor reserva.
  • El estudio destaca patrones específicos del sexo en el envejecimiento cerebral en contextos de enfermedades neurodegenerativas.
