  • La inflamación es un factor clave en la vulnerabilidad a enfermedades crónicas y la mortalidad en adultos mayores.
  • Se necesita investigación sobre los ritmos de sueño-vigilia y las citoquinas inflamatorias en el envejecimiento cerebral saludable.

Objetivo del estudio:

  • Investigar la asociación entre los patrones de sueño-vigilia y los marcadores inflamatorios en adultos mayores cognitivamente normales.
  • Caracterizar la relación entre las medidas objetivas del sueño y los perfiles de citoquinas.
  • Determinar si estas asociaciones son independientes de la patología de la enfermedad de Alzheimer.

Principales métodos:

  • 54 adultos mayores sanos se sometieron a actigrafía de 14 días y extracciones de sangre.
  • Se analizaron las citoquinas plasmáticas utilizando el panel de inflamación Alamar NULISASeq.
  • Se examinaron las asociaciones entre las métricas de actigrafía (eficiencia del sueño, duración, regularidad/variabilidad del ritmo) y 60 citoquinas seleccionadas mediante regresión lineal.

Principales resultados:

  • Un peor sueño y una alteración del ritmo circadiano se asociaron con un perfil elevado de citoquinas inflamatorias.
  • Una menor duración del sueño se relacionó con un mayor nivel de IL-1β; una peor eficiencia del sueño se relacionó con un mayor nivel de FGF23.
  • La regularidad del ritmo se asoció con IL-5RA; la fragmentación del ritmo con CSF2RB; una mayor amplitud con un menor nivel de IL-6.
  • Las asociaciones para IL-1β, IL-6, IL-5RA, FGF23 y CSF2RB se mantuvieron significativas después de controlar el p-tau217, lo que indica independencia de la patología de Alzheimer.

Conclusiones:

  • La alteración del sueño y los ritmos de 24 horas se asocian con un aumento de las citoquinas proinflamatorias y una disminución de las antiinflamatorias en adultos mayores sanos.
  • Estos hallazgos respaldan la hipótesis que conecta la alteración del sueño y la inflamación en el envejecimiento.
  • La investigación futura debe explorar el impacto de las interacciones sueño-inflamación en el envejecimiento cognitivo.
