Sus antecedentes:

  • La enfermedad de Alzheimer (EA) se asocia con el deterioro cognitivo, y factores genéticos como el alelo APOE4 aumentan el riesgo.; Las puntuaciones de riesgo poligénico (PRS) agregan variantes genéticas para predecir la susceptibilidad a la EA.; Este estudio investiga modelos de PRS en poblaciones brasileña (SABE) y europea (ADNI) para comprender las influencias genéticas en el deterioro cognitivo.

Objetivo del estudio:

  • Evaluar la efectividad de diferentes modelos de Puntuación de Riesgo Poligénico (PRS) en la predicción del deterioro cognitivo.; Examinar la influencia de la ascendencia genética y el alelo APOE4 en el rendimiento cognitivo en poblaciones mixtas y europeas.; Comparar el rendimiento de los modelos PRS en los conjuntos de datos SABE y ADNI.

Principales métodos:

  • Se generaron PRS utilizando PRSice-2, incluyendo los modelos Bel, Kunkle, Kunkle-AFR y PRS_SUM.; Se analizaron datos de las cohortes SABE (brasileña, n=1103) y ADNI (europea, n=785).; Se empleó regresión lineal con bootstrapping, controlando por edad, sexo, educación, estado de APOE4 y ascendencia local.

Principales resultados:

  • En SABE, la edad impactó negativamente la cognición, mientras que la educación la influyó positivamente. El APOE4 mostró un efecto menor, más pronunciado en individuos de ascendencia europea. Los modelos de PRS explicaron el 26.6-26.8% de la varianza.; El análisis de ascendencia local en SABE reveló una mayor homocigosidad en las ascendencias europea y de Asia oriental.; En ADNI, el APOE4 afectó significativamente las puntuaciones MMSE. El modelo Kunkle PRS (R²=0.155) mostró un mayor poder predictivo que Kunkle-AFR (R²=0.131) para la variabilidad cognitiva.

Conclusiones:

  • Los modelos PRS demuestran capacidades predictivas variables para el rendimiento cognitivo en diferentes poblaciones ancestrales.; La ascendencia local es crucial para comprender el deterioro cognitivo en poblaciones mixtas como la cohorte SABE.; Los factores genéticos, incluidos los PRS y el APOE4, interactúan de manera compleja con la ascendencia para influir en la salud cognitiva en las poblaciones que envejecen.
