  • Desentrañar los factores genéticos y ambientales en la etiología de la demencia ha sido un desafío.
  • Los estudios de gemelos ofrecen un enfoque único para controlar la confusión familiar en la investigación de la exposición laboral.

Objetivo del estudio:

  • Investigar la asociación entre las exposiciones laborales y el riesgo de demencia.
  • Evaluar el papel de la confusión familiar en la relación entre las exposiciones laborales y la demencia.
  • Examinar los tóxicos aéreos inhalables específicos y su impacto en el riesgo de demencia.

Principales métodos:

  • Estudio de cohorte longitudinal de gemelos (Swedish Adoption/Twin Study on Aging).
  • Exposiciones laborales autoinformadas evaluadas mediante cuestionario.
  • Análisis de Componentes Principales (PCA) para la exposición a múltiples contaminantes (PC1).
  • Modelos de Cox proporcionales y modelos de Cox estratificados para el análisis del riesgo de demencia.
  • Ajustes por edad, sexo, educación y tabaquismo.

Principales resultados:

  • La exposición laboral general (PC1) no mostró una asociación significativa con el riesgo de demencia.
  • La exposición a polvo, hollín y/o gases se asoció con un riesgo de demencia casi duplicado (HR 1.99).
  • Los análisis dentro de pares de gemelos revelaron una asociación comparable para polvo, hollín y/o gases (HR 2.30), lo que sugiere factores no familiares.

Conclusiones:

  • La exposición laboral al polvo, hollín y gases puede aumentar el riesgo de demencia.
  • La asociación observada probablemente no se explica por factores genéticos u otros factores familiares.
  • Se justifican más estudios a gran escala con datos de exposición detallados para confirmar estos hallazgos.
