Sus antecedentes:

  • Los estilos de vida sedentarios aceleran el envejecimiento, lo que provoca un declive en las funciones motora, metabólica y cognitiva.
  • La inflamación crónica, la disfunción mitocondrial y la resistencia a la insulina contribuyen al envejecimiento cerebral y a enfermedades neurodegenerativas como el Alzheimer y el Parkinson.
  • El ejercicio regular, incluido el entrenamiento aeróbico y de fuerza, puede mitigar estos declives relacionados con la edad y mejorar la plasticidad cerebral.

Objetivo del estudio:

  • Investigar los efectos del ejercicio aeróbico-de fuerza supervisado a corto plazo (3-9 meses) y a largo plazo (≥5 años) sobre la función cognitiva, la morfología cerebral y el metabolismo.
  • Identificar mediadores sistémicos, denominados exerkina, liberados por el músculo esquelético en respuesta al ejercicio.
  • Elucidar los mecanismos moleculares subyacentes a las respuestas adaptativas inducidas por el ejercicio a nivel sistémico, de órganos y molecular.

Principales métodos:

  • Ensayos clínicos aleatorizados (EMBRACE y AMETHYST) que involucran ejercicio aeróbico-de fuerza supervisado.
  • Resonancia magnética (RM) multimodal y espectroscopia para evaluar la estructura, función y metabolismo del cerebro.
  • Análisis de muestras biológicas (plasma, músculo, líquido cefalorraquídeo) para identificar exerkina y cambios metabólicos.
  • Evaluación de las funciones cognitivas, la aptitud física y la calidad de vida.

Principales resultados:

  • Se demostró la sostenibilidad y eficiencia de los programas de ejercicio a largo plazo en personas mayores.
  • Se identificó la regulación de moléculas bioactivas en el plasma y el líquido cefalorraquídeo en respuesta al ejercicio.
  • Se relacionaron los cambios moleculares inducidos por el ejercicio con el rendimiento cognitivo y el bienestar general.
  • Se estableció el papel de las exerkina secretadas por el músculo esquelético en la mediación de los beneficios del ejercicio.

Conclusiones:

  • El ejercicio regular de intensidad moderada a vigorosa, incluido el entrenamiento aeróbico y de fuerza combinado, es una estrategia potente para prevenir y tratar el deterioro cognitivo relacionado con la edad.
  • Las exerkina desempeñan un papel crucial en la mediación de los beneficios sistémicos del ejercicio.
  • Esta investigación proporciona una base para el desarrollo de prescripciones de ejercicio individualizadas para la práctica clínica.
