Sus antecedentes:

  • La enfermedad de Alzheimer (EA) es una causa principal de demencia, caracterizada por placas de amiloide-beta (Aβ) y ovillos de tau.
  • Las vesículas extracelulares (VE) son nanopartículas secretadas por células que pueden transportar biomoléculas e influir en la neuropatología.
  • El papel específico de las VE en los déficits conductuales relacionados con la EA sigue sin explorarse en gran medida.

Objetivo del estudio:

  • Investigar el impacto de las vesículas extracelulares (VE) derivadas del cerebro en la memoria y el comportamiento en un modelo de ratón.
  • Determinar si las VE de modelos de enfermedad de Alzheimer (EA) pueden inducir déficits cognitivos.

Principales métodos:

  • Se aislaron vesículas extracelulares (VE) de tejido cerebral post-mortem de pacientes de control y con EA, así como de ratones APP/PS1.
  • Estas VE se inyectaron estereotáxicamente en los hipocampos de ratones de tipo salvaje (WT).
  • La función cognitiva se evaluó mediante pruebas conductuales en varios momentos posteriores a la inyección.

Principales resultados:

  • Los ratones inyectados con VE de cerebros de control no mostraron alteraciones motoras ni conductuales.
  • Por el contrario, los ratones que recibieron VE de cerebros con EA exhibieron déficits de memoria significativos.
  • Estos déficits cognitivos fueron evidentes 14 y 21 días después de la inyección, lo que confirma que las VE de EA y las VE de APP/PS1 inducen la pérdida de memoria.

Conclusiones:

  • Las vesículas extracelulares (VE) pueden transmitir moléculas patológicas asociadas con la EA e inducir un deterioro de la memoria similar a la enfermedad.
  • Estos hallazgos resaltan las VE como un factor crítico en la patogénesis y progresión de la EA.
  • Las VE representan una diana terapéutica prometedora para enfermedades neurodegenerativas como el Alzheimer.
