Sus antecedentes:

  • El plexo coroideo (ChP) es crucial para la producción de líquido cefalorraquídeo (LCR) y el mantenimiento de la homeostasis del sistema nervioso central.
  • La disfunción del ChP se relaciona cada vez más con la patogénesis de las enfermedades neurodegenerativas.
  • Este estudio investiga la participación del ChP en la enfermedad de Alzheimer (EA), la enfermedad de Parkinson (EP) y la demencia con cuerpos de Lewy (DCL) utilizando proteómica del LCR.

Objetivo del estudio:

  • Explorar el papel de las proteínas expresadas en el plexo coroideo (ChP) en la fisiopatología de la EA, EP y DCL.
  • Identificar firmas proteicas específicas de la enfermedad en el LCR relacionadas con la función del ChP.
  • Comprender los mecanismos distintos que subyacen a estas enfermedades neurodegenerativas.

Principales métodos:

  • Análisis proteómico del LCR de 2902 proteínas en 150 pacientes con EA, 75 con EP, 53 con DCL y 47 controles.
  • Identificación de proteínas altamente expresadas en el ChP utilizando el Allen Brain Atlas.
  • Análisis de Redes de Coexpresión Genética Ponderada (WGCNA) para identificar módulos de proteínas y análisis de enriquecimiento de vías.

Principales resultados:

  • 799 de 2902 proteínas cuantificadas se expresaron en altos niveles en el ChP.
  • Las proteínas expresadas en el ChP desreguladas difirieron significativamente entre EA, EP y DCL.
  • WGCNA reveló módulos de proteínas distintos asociados con la inmunidad innata (disminuida en EA), procesos transcripcionales/traduccionales (aumentados en EP) y la matriz extracelular (aumentada en DCL).

Conclusiones:

  • El plexo coroideo (ChP) exhibe patrones de expresión proteica distintos en la EA, EP y DCL.
  • Estos hallazgos resaltan los roles específicos de la enfermedad de las proteínas expresadas en el ChP en la neurodegeneración.
  • Se necesita más investigación para aclarar la participación mecanicista del ChP en la progresión de la enfermedad.
